Overview
Acenocoumarol is an oral anticoagulant of the coumarin class used to prevent and treat thromboembolic disorders such as deep vein thrombosis, pulmonary embolism, atrial fibrillation-related stroke risk, and after heart valve replacement. It acts as a vitamin K antagonist by inhibiting the enzyme vitamin K epoxide reductase complex subunit 1 (VKORC1), thereby preventing the regeneration of reduced vitamin K. This inhibition reduces gamma-carboxylation and activation of vitamin K-dependent clotting factors II, VII, IX, and X as well as proteins C and S. The result is decreased blood coagulability[1][3][4][5][6]. Acenocoumarol is typically prescribed when warfarin is unsuitable or contraindicated for a patient[4].
