Acer macrophyllum pollen extract is an allergenic biological product derived from the pollen of the bigleaf maple tree, native to western North America. It is primarily utilized for the diagnosis and treatment of Type I hypersensitivity reactions, such as allergic rhinitis, allergic conjunctivitis, and allergic asthma, specifically those triggered by maple pollen exposure. As a diagnostic tool, it is used in percutaneous or intradermal skin testing to identify sensitization. As a therapeutic agent in allergen immunotherapy (AIT), it is administered in gradually increasing doses to induce immunological tolerance. The mechanism involves modulating the immune system to shift from a Th2-dominated allergic response to a Th1/Treg-mediated tolerant state, often characterized by the production of allergen-specific IgG4 blocking antibodies that compete with IgE for allergen binding.
