Acetaminophen + dexmedetomidine (BX-001) is an investigational fixed-dose combination analgesic developed for the management of moderate-to-severe acute post-operative pain. The formulation combines acetaminophen, a non-opioid analgesic that primarily inhibits prostaglandin synthesis in the central nervous system, with dexmedetomidine, a highly selective alpha-2 adrenergic receptor agonist. Dexmedetomidine provides analgesia and sedation by activating pre- and post-synaptic alpha-2 receptors in the locus coeruleus and spinal cord, inhibiting the release of norepinephrine and modulating pain transmission. By utilizing two agents with distinct, complementary mechanisms of action, the combination is designed to provide enhanced analgesic efficacy while potentially reducing the requirement for opioid medications and their associated side effects. The program was advanced into Phase 2 clinical trials by Baudax Bio, specifically targeting pain following orthopedic procedures such as bunionectomy.
