Acetram (DDS-06C) is an extended-release fixed-dose combination of the opioid analgesic tramadol hydrochloride and the non-opioid analgesic acetaminophen (paracetamol). Developed by Labopharm Inc. using its proprietary Contramid controlled-release technology, this formulation is designed to provide sustained pain relief over a 12-hour period, allowing for twice-daily dosing. Tramadol functions as a mu-opioid receptor agonist and a dual serotonin and norepinephrine reuptake inhibitor, while acetaminophen provides analgesic and antipyretic effects, likely through central cyclooxygenase inhibition and modulation of the endogenous cannabinoid system. This combination leverages the synergistic interaction between the two agents to manage moderate to moderately severe pain while potentially reducing the required dose of the opioid component. Although Labopharm faced regulatory challenges in the United States, the product received approval in several European markets.
