Drug intelligence / Profile preview
Vitamin K4 (acetomenaphthone) is a synthetic, water-soluble analog of vitamin K. It serves as a precursor to active vitamin K forms (such as vitamin K2) in the body, acting as an essential cofactor for gamma-glutamyl carboxylase (GGCX) to promote the post-translational carboxylation of clotting factors (factors II, VII, IX, and X) and anticoagulant proteins (proteins C and S). Clinically, it is approved for the treatment and prevention of hypoprothrombinemia and hemorrhage associated with vitamin K deficiency or malabsorption. Beyond its role in coagulation, acetomenaphthone has been investigated for its anti-inflammatory properties via the inhibition of the NLRP3 inflammasome, and its potential antineoplastic activity in various cancers, including prostate cancer, leukemia, and osteosarcoma. Recent preclinical research also highlights its potential in attenuating renal ischemia-reperfusion injury.
