Drug intelligence / Profile preview
Acetyl-6-formylpterin (Ac-6-FP) is a synthetic derivative of 6-formylpterin (6-FP), a photodegradation product of folic acid (vitamin B9). Ac-6-FP acts as a potent ligand and competitive antagonist of the major histocompatibility complex class I-related protein 1 (MR1). It binds covalently to MR1 via a Schiff base bond, stabilizing the protein and promoting its translocation to the cell surface. However, the resulting MR1-Ac-6-FP complex fails to engage and activate the T-cell receptor (TCR) of mucosal-associated invariant T (MAIT) cells, thereby inhibiting MAIT cell activation. Ac-6-FP is widely utilized as a pharmacological tool in preclinical research to investigate the role of the MR1-MAIT axis in various pathological conditions, including peritoneal fibrosis, bladder cancer, rheumatoid arthritis, and allergic asthma.
