Acetyl Dithranol is a small molecule derivative of dithranol (also known as anthralin), primarily used as a topical medication for the treatment of psoriasis. Its mechanism of action is multifaceted, involving the inhibition of keratinocyte hyperproliferation and DNA synthesis, which slows down the excessive cell division characteristic of psoriatic plaques. The drug accumulates in mitochondria, where it interferes with cellular energy supply, likely through the generation of free radicals. These free radicals are also implicated in the induction of lipid peroxidation. Additionally, Acetyl Dithranol has anti-inflammatory properties, inhibiting granulocyte function and the secretion of pro-inflammatory cytokines like IL-6, IL-8, and TNF-alpha by monocytes. It may also exert an immunosuppressive effect and reduce elevated cGMP levels. The drug was initially developed by the Shandong Institute Of Dermatology And Venereal Disease Control.
