Overview

Acetyl hexapeptide-8 (AH-8), commercially known as Argireline, is a synthetic peptide being investigated by the National Institute of Neurological Disorders and Stroke (NINDS) for the treatment of blepharospasm, a focal dystonia affecting the muscles that close the eye. It functions by mimicking the N-terminal end of the SNAP-25 protein, thereby competitively inhibiting the formation of the SNARE (Soluble NSF Attachment Protein Receptor) complex at the neuromuscular junction. This mechanism prevents the release of acetylcholine from synaptic vesicles, leading to localized muscle relaxation similar to the action of botulinum neurotoxin type A, but via a non-invasive topical application. In clinical trials, it has been evaluated as a topical cream to potentially extend the duration of botulinum toxin injections or serve as a standalone therapy for patients with blepharospasm.
