Overview

Acetyl plumbagin is a **synthetic small-molecule derivative of plumbagin**, a plant-derived naphthoquinone, that has been investigated in preclinical oncology studies as a **cholesterol-depleting anticancer agent**. In breast cancer cell models, particularly estrogen receptor-positive MCF-7 cells and tamoxifen-resistant models, acetyl plumbagin reduced intracellular cholesterol, lipid rafts, and cell viability, and enhanced apoptosis when combined with tamoxifen. Published studies also link its activity to modulation of cholesterol-homeostasis pathways and sensitization of breast cancer cells through CETP-associated biology, but a single direct molecular target has not been definitively established.
