Acetylated natural nucleotides is an investigational topical gel formulation being evaluated for the treatment of hand-foot syndrome (HFS), also known as palmar-plantar erythrodysesthesia (PPE). HFS is a common and painful skin toxicity associated with certain chemotherapy agents, most notably capecitabine. The treatment is designed to be applied topically to the palms and soles to mitigate the damage caused by anti-cancer medications. A Phase 2 clinical trial (NCT04592731), led by Nan Xu at Shanghai East Hospital, is investigating the efficacy and safety of this formulation in reducing the severity of HFS in cancer patients. The acetylation of the nucleotides likely enhances their lipophilicity and skin penetration, allowing them to support cellular repair processes in the affected dermal layers.
