Overview

ACH-000029 is a novel small molecule drug candidate developed for the treatment of anxiety disorders, including generalized anxiety disorder (GAD) and post-traumatic stress disorder (PTSD). It acts as a modulator at selected serotonergic and adrenergic receptors, specifically functioning as a partial agonist at 5-HT1A and 5-HT1D receptors, an antagonist at 5-HT2A receptors, and an antagonist at α1A-, α1B-, and α1D-adrenoceptors. Preclinical studies have demonstrated its anxiolytic-like effects in animal models of anxiety and PTSD, with efficacy comparable to established anxiolytics such as SSRIs (paroxetine, fluoxetine) and benzodiazepines. The compound shows good blood-brain barrier penetration and has not raised significant safety concerns in preclinical testing. Clinical development has reached phase 1 trials in healthy volunteers[1][3][7].
