Overview

ACH-1095 is a small molecule direct-acting antiviral (DAA) that targets the non-structural protein 5A (NS5A) of the hepatitis C virus (HCV). NS5A is a multifunctional phosphoprotein essential for the formation of the viral replication complex and the assembly of new infectious virus particles. Initially developed by Achillion Pharmaceuticals and later licensed to Gilead Sciences (where it was designated GS-9525), ACH-1095 was designed to provide potent, pan-genotypic inhibition of HCV replication. However, clinical development was discontinued as the therapeutic landscape for hepatitis C evolved toward highly effective, short-course combination regimens like sofosbuvir-based therapies, rendering earlier-generation single-agent candidates less competitive.
