AcHERV-HP16L1 is an experimental, non-replicating recombinant baculovirus-based DNA vaccine designed for the prevention and potential treatment of Human Papillomavirus (HPV) type 16 infections. The vaccine platform, termed AcHERV, utilizes an *Autographa californica* multiple nucleopolyhedrovirus (AcMNPV) vector that has been pseudotyped with the envelope protein of Human Endogenous Retrovirus (HERV) to facilitate efficient entry into human cells. AcHERV-HP16L1 carries the gene encoding the HPV 16 L1 major capsid protein. Upon intramuscular administration, the vector delivers the DNA into host cells, leading to the expression of the L1 protein, which triggers robust humoral immunity (neutralizing antibodies) and potent cell-mediated immune responses (IFN-gamma production). Preclinical studies in mice and pigs have demonstrated that AcHERV-HP16L1 provides high levels of immunogenicity and protection against HPV 16 pseudovirus challenge, often exceeding the cellular immune response elicited by conventional virus-like particle (VLP) vaccines.
