Overview

ACHN-IL2 is an experimental cell-based immunotherapy and gene therapy designed for the treatment of metastatic renal cell carcinoma (kidney cancer). It consists of allogeneic tumor cells derived from the ACHN human renal adenocarcinoma cell line that have been genetically modified to express and secrete interleukin-2 (IL-2). In clinical settings, ACHN-IL2 is typically administered via subcutaneous inoculation, often in combination with irradiated autologous tumor cells (CT-AUT). The mechanism of action involves the local production of IL-2 by the modified ACHN cells, which acts as a potent cytokine to stimulate the activation and proliferation of tumor-specific T-cells and natural killer (NK) cells. This localized immune stimulation is intended to overcome tumor-induced immunosuppression and mount a systemic anti-tumor response against the patient's own cancer cells. The therapy was primarily developed and evaluated in Phase 3 clinical trials by the Azienda Ospedaliera di Bologna Policlinico S. Orsola M. Malpighi in Italy.
