ACI-24 is a liposome-based active immunotherapy (vaccine) designed to stimulate the production of antibodies against aggregated beta amyloid (Aβ) peptides in the brain. Its mechanism involves presenting an array of truncated Aβ1–15 sequences anchored into liposomes, which adopt a β-sheet structure to form conformational epitopes that elicit a strong antibody response without activating pro-inflammatory T cells. The goal is to reduce or prevent the buildup of toxic amyloid plaques associated with Alzheimer's disease and Down syndrome-related dementia. Preclinical studies have shown that ACI-24 induces high titers of anti-Aβ antibodies and reduces amyloid pathology in animal models, while early clinical trials indicate good safety and tolerability profiles in humans[2][3][4][5]. The vaccine is being developed primarily by AC Immune, with Takeda as a major partner for late-stage development and commercialization[6][8].
