ACI-3024 is an investigational, oral small-molecule Tau aggregation inhibitor developed using AC Immune’s Morphomer technology, intended to treat tauopathies such as Alzheimer’s disease by stopping and reversing pathological Tau aggregation and reducing its spreading in the brain. A first-in-human randomized, placebo-controlled SAD/MAD Phase 1 study assessed safety, tolerability, pharmacokinetics, pharmacodynamics, CSF penetration, and a food effect; dosing in healthy volunteers and Alzheimer’s patients was included, with MRI assessments in multiple-ascending dose parts. The program originated from a collaboration and license between AC Immune and Eli Lilly to develop small-molecule Tau aggregation inhibitors for AD and other neurodegenerative diseases. Public databases subsequently list the asset as discontinued for Alzheimer’s disease, with highest global R&D status reported as discontinued. Mechanistically, it is categorized as a peptide aggregation inhibitor targeting Tau protein aggregates and positioned as a potential disease-modifying therapy for tau-driven neurodegeneration.[1][3][2][7][5]
