ACI-35.030 is an investigational active immunotherapy (vaccine) designed to generate a specific antibody response against pathological phosphorylated Tau (pTau) proteins in the brain, which are implicated in the development and progression of Alzheimer’s disease. The vaccine uses a liposomal formulation incorporating a conformationally-constrained, membrane-bound pTau peptide antigen along with adjuvants and T-helper peptides to stimulate the immune system. It aims to induce broad-spectrum protective antibodies that specifically target toxic forms of Tau while sparing normal endogenous Tau protein. Preclinical and early clinical studies have shown that ACI-35.030 elicits high titers of anti-pTau IgG antibodies, reduces tau pathology in animal models, and demonstrates safety as well as robust immunogenicity in humans. Developed using AC Immune’s SupraAntigen platform, it is currently being evaluated for its potential to slow or prevent disease progression by reducing pathological tau spreading during early stages of Alzheimer’s disease. The mechanism of action is to antagonize phosphorylated microtubule-associated protein tau (pTau).
