Acid-activatable curcumin polymer (ACP) is a therapeutic polymeric prodrug of curcumin. Curcumin, known for its potent antioxidant and anti-inflammatory activities, has low bioavailability, which ACP aims to overcome. In ACP, curcumin is covalently incorporated into the backbone of an amphiphilic polymer. This polymer self-assembles to form micelles that are designed to rapidly release curcumin under acidic conditions. ACP micelles have demonstrated potential as therapeutics for osteoarthritis by protecting articular structures from arthritis through the suppression of tumor necrosis factor-alpha (TNF-α) and interleukin 1β (IL-1β). Its pathological stimulus-responsiveness and potent activities suggest its potential for treating osteoarthritis and various other inflammatory diseases.
