acidic oligopeptide-DOTA-177Lu conjugate is a preclinical hydroxyapatite-targeted radioligand therapy designed for the treatment of bone metastases. Developed by researchers at Purdue University, the conjugate consists of an acidic oligopeptide ligand that selectively targets exposed hydroxyapatite in both osteolytic and osteoblastic bone lesions, tethered via a stable linker to a DOTA-chelated therapeutic radionuclide, lutetium-177 (177Lu). By homing specifically to exposed hydroxyapatite—which is normally occluded by a cartilaginous sheath in healthy bone—the therapy delivers localized beta radiation to destroy bone metastases while sparing healthy tissues and bone marrow.
