Acimtamig is a first-in-class tetravalent bispecific innate cell engager (ICE) designed to activate the innate immune system for the treatment of CD30-positive hematologic tumors. It is a recombinant antibody construct that binds to CD16A on natural killer (NK) cells and macrophages with two binding sites and to CD30 on tumor cells with the other two binding sites. This cross-linking brings NK cells into close proximity with tumor cells, enhancing NK cell-mediated antibody-dependent cellular cytotoxicity (ADCC) and leading to selective destruction of CD30-positive tumor cells. Acimtamig has shown promising efficacy in relapsed/refractory classical Hodgkin lymphoma and peripheral T-cell lymphoma, especially in combination with allogeneic NK cell therapies such as AlloNK. The drug was developed by Affimed Therapeutics in collaboration with The Leukemia & Lymphoma Society and University of Texas M.D. Anderson Cancer Center[1][2][3][4][5][7][8].
