Overview

AcJ8-D (also known as MJ8VAX or J8-DT) is an investigational peptide-based conjugate vaccine candidate developed by the Queensland Institute of Medical Research (QIMR Berghofer) for the prevention of Group A Streptococcus (GAS) infections and their post-infectious sequelae, such as acute rheumatic fever and rheumatic heart disease. The vaccine consists of an acetylated form of the J8 peptide—a minimal, non-host-reactive B-cell epitope derived from the highly conserved C-terminal region of the streptococcal M protein—chemically conjugated to a diphtheria toxoid (DT) carrier protein to provide T-cell help. Formulated with aluminum hydroxide (alum) as an adjuvant, AcJ8-D is designed to elicit broad-spectrum, protective, and opsonic antibody responses against multiple GAS serotypes without inducing autoimmune cross-reactivity. It has been evaluated in a Phase 1 clinical trial in healthy adult volunteers in Australia.
