Acoustic Cluster Therapy (ACT) is a novel ultrasound-mediated drug delivery platform developed by Exact-Therapeutics. The technology utilizes a unique formulation of microbubble-microdroplet clusters that are co-administered intravenously with standard therapeutic agents, such as chemotherapy. When targeted ultrasound is applied to the tumor site, these clusters undergo a phase shift and expand into larger bubbles that oscillate, inducing biomechanical effects on the tumor vasculature and cell membranes, a process known as sonoporation. This localized activation significantly enhances the extravasation and penetration of the co-administered drug into the tumor tissue, potentially increasing therapeutic efficacy while reducing systemic side effects. ACT is currently being evaluated in clinical trials, including the ACTIVATE study for patients with colorectal liver metastases and pancreatic cancer.
