ACP-017 is an investigational, intratumorally administered prodrug of a Toll-like receptor 7/8 (TLR7/8) agonist, developed by Ascendis Pharma using its proprietary TransCon (Transient Conjugation) technology. The TransCon platform involves a carrier (typically a methoxy polyethylene glycol) and a unique linker that allows for the sustained, local release of the active, small-molecule TLR7/8 agonist directly within the tumor microenvironment. By activating TLR7 and TLR8 on innate immune cells such as dendritic cells and macrophages, ACP-017 aims to stimulate a potent local inflammatory response that subsequently triggers a systemic, T-cell-mediated anti-tumor immune response (the abscopal effect). This localized delivery approach is designed to maximize therapeutic efficacy while minimizing the systemic cytokine release and toxicity often associated with systemic TLR agonists. ACP-017 is currently being evaluated in Phase 1/2 clinical trials for various solid tumors, including HPV-associated cancers and cutaneous malignancies, both as a monotherapy and in combination with PD-1 checkpoint inhibitors.
