Overview

ACR246 is an investigational antibody-drug conjugate (ADC) developed by Hangzhou Adcoris Biopharmacy. It targets the 5T4 transmembrane glycoprotein, which is overexpressed in a variety of solid tumors and associated with increased tumor invasiveness. The ADC consists of a humanized monoclonal antibody directed against 5T4, site-specifically conjugated to D2102, a novel DNA topoisomerase I inhibitor payload, via a stable and tumor microenvironment-cleavable linker. Upon binding to 5T4-positive tumor cells, ACR246 is internalized and releases D2102 intracellularly, inducing DNA damage, cell cycle arrest, and apoptosis specifically in cancer cells. Preclinical studies have demonstrated robust anti-tumor activity across multiple solid tumor models with favorable safety profiles. ACR246 is currently being evaluated in Phase 1/2 clinical trials for advanced solid tumors[1][2][3][4][5].
