Overview

ACR317 is a novel antibody-drug conjugate (ADC) targeting Cadherin-17 (CDH17), developed by Adcoris Biopharma for the treatment of gastrointestinal (GI) cancers, including colorectal and gastric cancer. The ADC utilizes a fully human antibody, IBR217, which is site-specifically conjugated to a novel topoisomerase I inhibitor payload via the proprietary MuSC™ platform, achieving a drug-to-antibody ratio (DAR) of 8. CDH17 is a validated target with high expression in GI malignancies and restricted expression in normal tissues, primarily on the basolateral membrane of intestinal epithelial cells. Preclinical data presented at AACR 2026 demonstrated that ACR317 has high affinity and specificity for CDH17, efficient internalization, and potent antitumor activity in multiple cell-derived xenograft (CDX) models, outperforming benchmark ADCs.
