ACR335 is a first-in-class bispecific antibody-drug conjugate (BsADC) targeting cMET and EGFR, developed by Hangzhou Adcoris Biopharma. It is constructed using the proprietary MuSC™ platform, which enables the site-specific conjugation of two distinct cytotoxic payloads to the bispecific antibody IBR335. The payloads consist of a Topoisomerase I (Top1) inhibitor and a second, non-Top/non-Tubulin inhibitor, achieving a total drug-to-antibody ratio (DAR) of 8 (4+4). This dual-payload strategy is designed to synergistically enhance tumor cell killing and mitigate therapeutic resistance driven by pathway crosstalk between cMET and EGFR. Preclinical data demonstrate potent antitumor activity and regression in various solid tumor models, including gastric cancer, non-small cell lung cancer (NSCLC), and bladder cancer. Phase I clinical trials are anticipated to begin in the second quarter of 2026.
