Acrizanib is a small molecule tyrosine kinase inhibitor that selectively targets vascular endothelial growth factor receptor 2 (VEGFR2). It is being investigated as a treatment for neovascular (wet) age-related macular degeneration and other ocular diseases characterized by pathological angiogenesis. Acrizanib works by binding to the intracellular domain of VEGFR2, inhibiting its phosphorylation and downstream signaling pathways, thereby suppressing abnormal blood vessel formation in the eye. Preclinical studies have shown that intravitreal injection of acrizanib reduces pathological neovascularization, inflammation, and vascular leakage without significantly affecting normal physiological angiogenesis or retinal thickness. The drug has completed Phase 2 clinical trials for neovascular age-related macular degeneration[1][3][4][5][7].
