Acrizanib (LHA510) is a potent, small-molecule tyrosine kinase inhibitor developed by Alcon (a Novartis company) for the treatment of exudative (wet) age-related macular degeneration (nAMD). It primarily targets the vascular endothelial growth factor receptor (VEGFR) family, with high selectivity for VEGFR-2. The drug was investigated as a topical ophthalmic solution intended for maintenance therapy to reduce the frequency of intravitreal anti-VEGF injections, such as ranibizumab. By inhibiting VEGFR signaling, acrizanib aims to suppress the pathological angiogenesis and vascular permeability that drive the progression of wet AMD. However, clinical development for this indication was discontinued after Phase 2 trials failed to demonstrate a significant reduction in the need for supplemental injections compared to the control group.
