ACS2015 is a non-viral, gene-modified chimeric antigen receptor T-cell (CAR-T) therapy targeting the Ephrin type-B receptor 4 (EPHB4). Developed by A-Seeds Australia (a spin-out from Shinshu University), the therapy utilizes the piggyBac transposon system for stable gene delivery, which avoids the use of viral vectors and may reduce T-cell exhaustion. The CAR construct incorporates a modified version of the natural EPHB4 ligand, ephrin B2, as its binding domain. ACS2015 is currently being evaluated in Phase I/Ib clinical trials for the treatment of EPHB4-expressing solid tumors, including colorectal cancer, hepatocellular carcinoma, and various sarcomas.
