Drug intelligence / Profile preview
ACT-1014-6470 is a novel, orally available small molecule antagonist of the complement factor 5a receptor 1 (C5aR1). It was developed for anti-inflammatory activity, specifically targeting C5a-mediated effects in the complement system without interfering with C5b-mediated membrane attack complex (MAC) formation, thereby preserving bactericidal activity. The drug is being investigated as a potential treatment for immune-mediated and autoimmune disorders. In Phase 1 clinical studies, ACT-1014-6470 demonstrated potent inhibition of C5aR1 activation in preclinical models and was well tolerated in healthy volunteers[1][2][3][4][5]. The compound shows dose-proportional pharmacokinetics and sustained target engagement[5].
