Overview

ACT-1239 is a synthetic, pre-erythrocytic malaria vaccine candidate developed by Artificial Cell Technologies (ACT). It is constructed using ACT's proprietary layer-by-layer microparticle (LbL-MP) platform, which involves the assembly of ultra-thin polypeptide nanofilms onto particulate cores. The vaccine targets the circumsporozoite (CS) protein of *Plasmodium falciparum*, the primary parasite causing human malaria. ACT-1239 is designed to be self-adjuvanted through the incorporation of a TLR-2 agonist (Pam3) within the particle structure, aiming to induce both sporozoite-neutralizing antibodies and Th1-biased T-cell responses. A Phase 1a clinical trial in Australia was completed in 2021, demonstrating that the vaccine was safe, tolerable, and immunogenic in healthy participants.
