Overview

ACT-462206 is a potent, selective, orally bioavailable dual orexin receptor antagonist that inhibits the stimulating effects of orexin peptides at both the orexin 1 and orexin 2 receptors. It was developed as a potential treatment for insomnia and demonstrated dose-dependent sedative effects in human trials, with sleepiness and headache being the most common adverse events. In preclinical studies, it showed anxiolytic-like properties without affecting cognition or motor function. The drug was originally developed by Actelion Pharmaceuticals but its clinical development for insomnia was discontinued after phase I trials[1][2][3][5][6].
