Overview

ACT-774312 is an orally bioavailable, selective antagonist of the chemoattractant receptor-homologous molecule expressed on T-helper 2 cells (CRTH2; also known as prostaglandin D2 receptor 2 or G-protein coupled receptor 44). It is being developed primarily for the treatment of nasal polyposis and other allergic or type-2 inflammatory diseases. By blocking CRTH2, ACT-774312 inhibits the action of prostaglandin D2 on Th2 cells, which may reduce allergic and inflammatory responses. The drug has demonstrated dose-proportional pharmacokinetics with a terminal elimination half-life of about 12 hours and was well tolerated in clinical studies. The primary developers are Actelion Pharmaceuticals and Idorsia Pharmaceuticals[1][3][5].
