ACT-903 is an antibody-drug conjugate (ADC) composed of recombinant human alpha-fetoprotein (AFP) linked to a proprietary maytansinoid cytotoxic payload. The drug is designed to selectively target alpha-fetoprotein receptor, which are found on the majority of solid and liquid cancer cells (>80%) but not on healthy adult cells except for myeloid-derived suppressor cells (MDSCs). By leveraging AFP as a targeting shuttle, ACT-903 delivers its cytotoxic payload directly to cancer and immune suppressor cells, aiming for high efficacy with minimal toxicity. Preclinical studies have demonstrated significant anti-tumor activity and excellent safety in multiple xenograft models, including colorectal and ovarian cancers. The drug has shown tumor regression, increased survival rates, and no observable toxicity in animal models. ACT-903 is being developed by Alpha Cancer Technologies for indications such as platinum-resistant ovarian cancer and testicular germ cell tumors, with plans to seek Orphan Drug or Breakthrough Therapy Designation.
