Overview

ACTH4-10Pro8-Gly9-Pro10 is a synthetic peptide analogue derived from the adrenocorticotropic hormone (ACTH), specifically corresponding to amino acids 4 through 10 with substitutions at positions 8 (proline), 9 (glycine), and 10 (proline). It is devoid of classical hormonal effects but exhibits neuromodulatory and neuroprotective properties. The drug has demonstrated anti-inflammatory effects by reducing pro-inflammatory cytokines such as IL-6 and IL-8, while increasing anti-apoptotic proteins like Bcl-2 and neurotrophic factors such as BDNF. These actions are associated with reduced neuronal apoptosis, enhanced neuronal survival, improved recovery in models of traumatic brain injury, spinal cord injury, stroke, and Alzheimer’s disease. The compound can be administered intranasally for rapid onset[2][3][6].
