Actinium-225-labeled daratumumab is an experimental antibody radiation-conjugate (ARC) being developed by Actinium Pharmaceuticals as part of its AWE (Actinium Warhead Enabling) Technology Platform. This radiopharmaceutical combines the monoclonal antibody daratumumab, which targets the CD38 antigen, with the potent alpha-particle emitting radioisotope actinium-225. CD38 is a glycoprotein highly expressed on the surface of multiple myeloma cells and certain lymphoma cells, making it an effective target for directed therapy. By delivering high-energy alpha radiation directly to the tumor cells, the conjugate induces lethal double-stranded DNA breaks, significantly enhancing the cytotoxic effect compared to the unlabeled antibody while preserving the antibody's targeting specificity and Fc-mediated effector functions. The compound is currently in preclinical development for the treatment of multiple myeloma and lymphoma.
