Drug intelligence / Profile preview
Actinium-225-labeled titanium dioxide nanoparticles ([225Ac]TiO2) are radiolabeled metal oxide nanoparticles being investigated as potential carriers for targeted alpha therapy (TAT) in oncology. Titanium dioxide (TiO2) nanoparticles are utilized as a carrier matrix to stably retain the alpha-emitting radionuclide actinium-225 (225Ac) and its radioactive decay progeny (such as francium-221 and bismuth-213). This retention is critical to prevent the escape of daughter radionuclides (recoil effect) and minimize off-target toxicity to healthy tissues. The nanoparticles are prepared by hydrolysis of tetra-n-butyl orthotitanate and labeled with 225Ac via surface sorption or doping during synthesis. They are being evaluated in preclinical research for their stability, progeny retention, and biodistribution.
