Actinomycin D (also known as dactinomycin) is a cytotoxic antibiotic and antineoplastic agent derived from Streptomyces parvullus. It is used primarily in the treatment of various cancers, including Wilms' tumor, rhabdomyosarcoma, Ewing's sarcoma, gestational trophoblastic neoplasia (including malignant hydatidiform mole), testicular cancer (metastatic nonseminomatous), and certain types of ovarian cancer. Actinomycin D works by binding to DNA and inhibiting RNA synthesis (transcription), particularly affecting chain elongation. This inhibition leads to decreased mRNA production and subsequent reduction in protein synthesis within cells. The drug is administered intravenously and is highly toxic; it must be handled with care due to its corrosive properties and risk of severe tissue damage if extravasation occurs[1][3][5][6].
