Overview

Activated haploidentical allogeneic NK cells (NKAE cells) are an investigational adoptive cell immunotherapy. The therapy involves the ex vivo activation and expansion of natural killer (NK) cells obtained from a haploidentical (half-matched) donor. These cells are then infused into patients, typically following lymphodepleting chemotherapy or low-dose radiotherapy, to enhance their anti-tumor activity. In clinical studies led by Antonio Pérez-Martínez at the Hospital Universitario La Paz, the cells are administered alongside interleukin-2 (IL-2) to treat pediatric and young adult patients with refractory sarcomas, including Ewing sarcoma, rhabdomyosarcoma, and osteosarcoma. The mechanism relies on the innate ability of NK cells to recognize and kill tumor cells through various receptors, bypassing the need for MHC-restricted antigen presentation.
