Overview
An activin A neutralizing antibody is a monoclonal antibody designed to bind and neutralize activin A, a member of the transforming growth factor-beta (TGF-β) superfamily. Activin A is a homodimer of the inhibin beta A chain (INHBA) and plays a key role in various physiological and pathological processes, including tissue fibrosis, inflammation, and bone remodeling. In chronic kidney disease (CKD), activin A is induced by TGF-β1 and acts as a critical mediator of both glomerular and tubulointerstitial fibrosis. Neutralization of activin A has been shown to inhibit Smad3 activation and mitigate renal fibrosis in preclinical models. Additionally, activin A neutralizing antibodies (such as garetosmab) are being investigated for the treatment of fibrodysplasia ossificans progressiva (FOP), where mutant ACVR1 is aberrantly activated by activin A to drive heterotopic ossification.
