Overview

ACTM-838 is a highly attenuated, precision genome-engineered bacterial immunotherapy developed by Actym Therapeutics for the treatment of advanced solid tumors. It utilizes the STACT platform to deliver two immune-modulatory payloads—an engineered IL-15 superagonist complex (IL-15plex, composed of IL-15 and IL-15Ra) and a constitutively active engineered STING (eSTING)—directly to tumor-resident phagocytic antigen-presenting cells within the tumor microenvironment. The therapy is designed to reprogram the local immune state from an immunosuppressive phenotype to an anti-tumor phenotype by activating both innate and adaptive immunity. Preclinical studies have shown that ACTM-838 can eradicate solid tumors, induce durable anti-tumor memory responses, and promote CD8+ T-cell mediated tumor clearance even in T-cell excluded tumors. The drug is currently being evaluated in a Phase 1a/1b clinical trial for patients with advanced solid tumors who have failed prior therapies.
