**ACTR087** is an investigational autologous engineered T-cell therapy developed by Unum Therapeutics, subsequently renamed Cogent Biosciences. Patient-derived T cells are transduced ex vivo to express an Antibody-Coupled T-cell Receptor comprising a CD16V158 extracellular Fc-binding domain linked to 4-1BB and CD3ζ signaling domains. The engineered cells bind the Fc region of a co-administered tumor-targeting IgG antibody, allowing the antibody to determine tumor-antigen specificity and triggering T-cell activation and cytotoxicity. ACTR087 was evaluated in Phase 1 studies with rituximab for relapsed or refractory CD20-positive B-cell lymphomas and with SEA-BCMA for relapsed or refractory multiple myeloma. The lymphoma program was deprioritized after safety-related regulatory events, and the multiple-myeloma study was terminated.
