Overview

ACTR087 + rituximab is an investigational combination therapy that consists of autologous T lymphocytes genetically modified to express the "Antibody Coupled T-cell Receptor" (ACTR087—a universal chimeric receptor incorporating CD16V158, CD3ζ, and 4-1BB signaling domains), used together with rituximab, an anti-CD20 monoclonal antibody. The ACTR087-modified T cells engage the Fc region of rituximab bound to CD20-positive B cells, promoting targeted tumor cell killing via antibody-dependent cell-mediated cytotoxicity and T cell activation. This approach is designed to both enhance antitumor immune responses and increase the specificity of T-cell cytotoxicity for tumors targeted by therapeutic antibodies. Developed primarily for relapsed/refractory CD20-positive B cell lymphoma, it has also been explored preclinically in other antibody-directed settings.
