ACU2086 is a synthetic, non-cyclic dinucleotide small molecule agonist of the Stimulator of Interferon Genes (STING) pathway. Developed by Aculeus Therapeutics, it is designed for systemic delivery to treat hematological malignancies, specifically acute myeloid leukemia (AML). ACU2086 exerts potent cell-intrinsic anti-leukemic activity by inducing STING-dependent apoptosis through BAX and BAK, notably maintaining efficacy in high-risk AML cases with TP53 mutations. Preclinical studies demonstrate that ACU2086 acts synergistically with the BCL-2 inhibitor venetoclax, significantly reducing leukemic burden in vivo.
