ACX-01 is an investigational immunotherapy candidate being developed by Centauri Therapeutics for the treatment of colorectal cancer and head and neck cancer. It is built upon the company's proprietary Alphamer platform, which uses a modular approach to redirect the immune system. ACX-01 consists of a targeting moiety—either a full-length monoclonal antibody or a Fab fragment—conjugated to an alpha-Gal epitope. This carbohydrate epitope is naturally recognized by high titers of pre-existing anti-Gal antibodies in the human bloodstream. By binding to tumor-associated antigens on the surface of cancer cells, ACX-01 recruits these anti-Gal antibodies to the tumor site, subsequently triggering potent effector functions such as complement-dependent cytotoxicity (CDC) and antibody-dependent cellular cytotoxicity (ADCC) to eliminate the target cells.
