Overview

**AD-206** is an Addpharma fixed-dose oral combination of **esomeprazole** and **calcium carbonate**. In the evaluated formulations, it contained esomeprazole 20 mg or 40 mg plus calcium carbonate 600 mg. Esomeprazole is a proton-pump inhibitor that suppresses gastric acid secretion through inhibition of gastric hydrogen-potassium ATPase, while calcium carbonate provides antacid activity and raises gastric pH. In a completed randomized Phase 1 crossover study in healthy male volunteers, AD-206 produced comparable multiple-dose esomeprazole exposure and gastric-acid suppression versus conventional esomeprazole, with faster time to peak esomeprazole concentration and faster attainment of intragastric pH 4.
