AD-231C is a next-generation antibody-drug conjugate (ADC) targeting Receptor Tyrosine Kinase Like Orphan Receptor 1 (ROR1), a protein frequently overexpressed in a variety of solid and hematological malignancies. Developed by Adcentrx Therapeutics using its proprietary i-DEX platform, AD-231C consists of a humanized anti-ROR1 monoclonal antibody conjugated to a potent topoisomerase I inhibitor payload through a stable, yet selectively cleavable linker. Upon binding to ROR1 on the tumor cell surface, the ADC is internalized, and the payload is released to induce DNA damage and subsequent apoptosis. The i-DEX platform is designed to enhance the stability of the ADC in circulation while ensuring efficient payload release within the tumor microenvironment, potentially offering an improved safety profile and enhanced efficacy compared to earlier ROR1-targeting candidates. AD-231C is currently undergoing Phase 1 clinical evaluation for the treatment of advanced solid tumors.
