Overview

Ad-4-1BBL is a recombinant adenoviral vector engineered to express the 4-1BB ligand (4-1BBL, also known as CD137L or TNFSF9). 4-1BBL is a potent costimulatory molecule that binds to the 4-1BB (CD137) receptor, primarily expressed on activated T cells and natural killer (NK) cells. By delivering the 4-1BBL gene via an adenovirus, this therapy aims to enhance the immune response against tumors by promoting T-cell proliferation, cytokine production, and cytotoxic activity. It has been studied in the context of modifying dendritic cells to improve their ability to prime T-cell responses and as a direct intratumoral injection to stimulate the local microenvironment. The vector is typically based on a replication-deficient adenovirus (such as Ad5) to ensure safety while providing high levels of transient gene expression.
