Overview

Ad AFP D55-TRAIL is a gene therapy product consisting of a recombinant oncolytic adenovirus engineered for enhanced liver cancer targeting. It utilizes an α-fetoprotein (AFP)-regulated promoter and deletes the E1B-55K region (D55) to confer selective replication in AFP-positive liver cancer cells. The virus is further armed with the human gene coding for tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), aiming to induce apoptosis specifically in malignant cells. The therapy leverages cancer-targeting gene-viro-therapy, delivering TRAIL through the selectively replicating adenovirus to maximize tumor cell apoptosis while minimizing effects on normal tissue. This approach was initially developed by the Shanghai Institutes for Biological Sciences[1][5].
