Ad-AR-V7 is an adenoviral-based cancer vaccine designed to target the androgen receptor splice variant-7 (AR-V7), a constitutively active form of the androgen receptor that lacks the ligand-binding domain and is frequently associated with resistance to androgen deprivation therapy in castration-resistant prostate cancer (CRPC). Developed by Herbert Kim Lyerly and his team at Duke University, the vaccine utilizes an adenoviral vector platform to deliver and express AR-V7-derived neoepitopes, primarily targeting the N-terminus of the protein. This approach is intended to elicit a robust, antigen-specific T cell response against tumor cells expressing the splice variant. Preclinical evidence suggests that Ad-AR-V7 can suppress tumor growth and improve survival, with enhanced efficacy observed when used in combination with androgen receptor antagonists like enzalutamide or immune checkpoint inhibitors such as anti-PD-1 antibodies.
